Determination of Phamacophoric Groups

exemplifying the effect of the STD NMR by removing all peaks of nonbinding components
from the spectrum leaving only the signals of the binding epitope.
Testing for binding activity with STD NMR is very fast. It can be estimated
that with STD NMR more than 50.000 compounds can be screened per day for binding
activity. This allows to use NMR as a HTS screening protocol.
STD NMR is perfectly well suited to investigate membrane bound proteins. This
is of central importance because many pharmaceutically important targets are
membrane bound proteins. Usually, these proteins loose their activity upon isolation
from the membrane environment. With STD NMR these proteins can be investigated
in their natural environment, the membrane. It is very important to note that
there is no size limit for the protein used.
